It was not actually settlers who wanted to go first, or who were prepared to suffer being in the wilderness. It was the evangelists (missionaries).
The Whitmans believed they had been told to go west and take the Word of God to the Native Americans.
So it was the evangelists who first made the journey west, taking a route that would eventually be the Oregon Trail. In fact the Whitman Mission was stop along the Oregon Trail route.
